痛风性关节炎(GA)是一种痛感强烈的炎症。Luminex液相细胞因子芯片帮助研究者探讨感觉神经元神经肽和趋化因子信号对炎症诱导巨噬细胞极化的影响。
首先研究人员通过RNA-seq 筛选出神经性关节炎中关键靶点基因sFRP2,并通过体内外实验验证sFRP2的功能,沉默sFRP2可显著抑制炎性细胞浸润减轻痛觉。 通过运用Luminex液相细胞因子芯片技术,研究人员发现sFRP2促进骨髓来源的巨噬细胞(BMM)向M1极化和迁移,上调了促炎细胞因子,参与炎症反应。 最后研究人员确认了背根神经节神经元(DRG)释放的分泌蛋白sFRP2通过调节Wnt/β-catenin和NF-κB信号通路,促进BMM向M1极化和迁移,进而参与炎症反应。而表达量上调的促炎细胞因子又可以诱导内皮细胞凋亡。 研究证实了感觉神经-巨噬细胞在GA病理中的作用,通过旁分泌方式调控了sFRP2的表达。促进NF-κB信号通路的激活,引起了炎症反应。为今后GA在痛风炎症缓解和治疗方面的研究提供了新的视角。

卵巢癌是女性最致命的恶性肿瘤,肿瘤的复发和转移使得其治疗具有难度。研究人员利用Luminex液相细胞因子芯片研究发现顺铂刺激的经典活化巨噬细胞(CAMs)通过增加CCL20的产生来促进卵巢癌细胞的迁移,揭示了一种新的顺铂和CAMs相互作用驱动的肿瘤细胞迁移机制。
首先,研究人员选用Luminex液相细胞因子芯片对顺铂处理的THP-1和PBMC细胞的上清进行细胞因子筛选,证明顺铂刺激的经典活化巨噬细胞(CAMs)通过上调CCL20的表达来进一步激活卵巢癌细胞上的CCR6受体,触发上皮向间充质的转化。当CCR6沉默阻断后,CAMs对肿瘤细胞的促迁移作用受到抑制。 同时,研究人员在卵巢癌临床组织标本中,也发现了卵巢癌组织中高表达CCR6的患者生存时间明显降低。 最终研究人员揭示了一种新的由顺铂和CAMs间串扰驱动的肿瘤细胞迁移机制,并提示CCL20-CCR6可能是降低晚期卵巢癌化疗诱导转移的潜在治疗靶点。
